    Application of chicken microarrays for gene expression analysis in other avian species

    BACKGROUND: With the threat of emerging infectious diseases such as avian influenza, whose natural hosts are thought to be a variety of wild water birds including duck, we are armed with very few genomic resources to investigate large scale immunological gene expression studies in avian species. Multiple options exist for conducting large gene expression studies in chickens and in this study we explore the feasibility of using one of these tools to investigate gene expression in other avian species. RESULTS: In this study we utilised a whole genome long oligonucleotide chicken microarray to assess the utility of cross species hybridisation (CSH). We successfully hybridised a number of different avian species to this array, obtaining reliable signals. We were able to distinguish ducks that were infected with avian influenza from uninfected ducks using this microarray platform. In addition, we were able to detect known chicken immunological genes in all of the hybridised avian species. CONCLUSION: Cross species hybridisation using long oligonucleotide microarrays is a powerful tool to study the immune response in avian species with little available genomic information. The present study validated the use of the whole genome long oligonucleotide chicken microarray to investigate gene expression in a range of avian species

    Effects of Microstructure Formation on the Stability of Vapor Deposited Glasses

    Glasses formed by physical vapor deposition (PVD) are an interesting new class of materials, exhibiting properties thought to be equivalent to those of glasses aged for thousands of years. Exerting control over the structure and properties of PVD glasses formed with different types of glass-forming molecules is now an emerging challenge. In this work, we study coarse grained models of organic glass formers containing fluorocarbon tails of increasing length, corresponding to an increased tendency to form microstructures. We use simulated PVD to examine how the presence of the microphase separated domains in the supercooled liquid influences the ability to form stable glasses. This model suggests that increasing molecule tail length results in decreased thermodynamic and kinetic stability of the molecules in PVD films. The reduced stability is further linked to the reduced ability of these molecules to equilibrate at the free surface during PVD. We find that as the tail length is increased, the relaxation time near the surface of the supercooled equilibrium liquid films of these molecules are slowed and become essentially bulk-like, due to the segregation of the fluorocarbon tails to the free surface. Surface diffusion is also markedly reduced due to clustering of the molecules at the surface. Based on these results, we propose a trapping mechanism where tails are unable to move between local phase separated domains on the relevant deposition time scales

However, there is a trend to reduce this use of antibiotics in feedstuffs, and legislation is now in place in Europe to prohibit their use in this way. As a consequence, economically important diseases, such as necrotic enteritis (NE) of chickens, that are caused by Clostridium perfringens have become more prevalent. Recent research is creating a paradigm shift in our understanding of the pathogenesis of NE and is now providing information that will be necessary to monitor and control the incidence of NE in poultry

    Beliefs About Consciousness and Reality of Participants at ‘Tucson II’

    In previous studies we had found correlations between the material-transcendent dimension underlying the Western intellectual tradition and the diversity of ideas concerning consciousness. In the course of our work we developed the Beliefs About Consciousness and Reality Questionnaire that could be used for measuring fundamental beliefs about consciousness and reality. A survey of participants at the scientific meeting Toward a Science of Consciousness 1996 “Tucson II” was conducted using this questionnaire. Results from 212 respondents indicated scores substantially in the transcendent direction, both for scales underlying the questionnaire as well as for some of its individual items, relative to a 1986 standardization sample. Having traditional or one’s own religious beliefs, interest in phenomenology and culture, lack of interest in neural correlates and age were also all correlated with scale scores in the transcendent direction. Given the diversity of fundamental beliefs about consciousness and reality of listeners at a meeting such as Tucson II, speakers need to find ways to communicate across the spectrum of the material-transcendent dimension

    Weekly Versus Monthly Testosterone Administration On Fast and Slow Skeletal Muscle Fibers in Older Adult Males

    Context: In older adults, loss of mobility due to sarcopenia is exacerbated in men with low serum T. T replacement therapy is known to increase muscle mass and strength, but the effect of weekly (WK) vs monthly (MO) administration on specific fiber types is unknown. Objective: To determine the efficacy of WK vs MO T replacement on the size and functional capacity of individual fast and slow skeletal muscle fiber types. Design, Setting, and Patients: Subjects were randomized into a 5-month, double-blind, placebo-controlled trial. All subjects (ages, 61–71 y) were community-dwelling men who had T levels \u3c 500 ng/dL. Intervention: Subjects were dosed weekly for 5 months, receiving continuous T (WK, n = 5; 100 mg T enanthate, im injection), monthly cycled T (MO, n = 7; alternating months of T and placebo), or placebo (n = 7). Muscle biopsies of the vastus lateralis were obtained before and after treatment. Main Outcome Measures: Main outcomes for individual slow and fast fibers included fiber diameter, peak force (P0), rate of tension development, maximal shortening velocity, peak power, and Ca2+ sensitivity. Results: Both treatments increased fiber diameter and peak power, with WK treatment 5-fold more effective than MO in increasing type I fiber P0. WK effects on fiber diameter and force were 1.5-fold higher in slow fibers compared to fast fibers. In fast type II fibers, diameter and P0 increased similarly between treatments. The increased power was entirely due to increased fiber size and force. Conclusions: In conclusion, T replacement effects were fiber-type dependent, restricted to increases in cell size, P0, and peak power, and dependent on the paradigm selected (WK vs MO)
